> Light sheet microscopy (LSM) is an essential tool for quantitative imaging of cellular dynamics in the embryonic development of complex organisms such as Drosophila and zebrafish. Multiple images from different viewpoints over long periods of time are recorded to obtain optimal coverage of the specimen in space and time, generating terabytes of three-dimensional (3D) image stacks. These time-lapse recordings contain all the necessary information to reconstruct complete cell lineages during long periods of time (over 24 hours) over the entire developing embryo. However, there are many technical challenges to achieve this goal. In this talk I will present an introduction to LSM and an overview of the SiMView technology developed in Keller Lab at Janelia Farm Research Campus. The framework is a comprehensive high-throughput pipeline to efficiently acquire, process and analyze LSM recordings of developing organisms.
