ABSTRACT:

Aggregation of Aβ peptides is related to the onset of Alzheimer’s disease, but the molecular mechanism of Aβ fibril formation is still poorly understood. Thermodynamics and energetics of the deposition of Aβ monomers on the preformed amyloid fibril were probed by replica exchange molecular dynamics with implicit solvent model. Consistent with the experiments the results showed that the fibril growth follows the dock-lock mechanism. The interactions that stabilize the binding of the peptide to the fibril and the preferable binding region in the peptides were identified. Also, a single-site computational mutation effects on the fibril growth were investigated. The results indicated that the strong off-registry side chain interactions may impede fibril growth.
